Descriptive evaluation of antibody responses to severe acute respiratory coronavirus virus 2 (SARS-CoV-2) infection in plasma and gingival crevicular fluid in a nursing home cohort—Arkansas, June–August 2020

Objective:
To characterize and compare severe acute respiratory coronavirus virus 2 (SARS-CoV-2)–specific immune responses in plasma and gingival crevicular fluid (GCF) from nursing home residents during and after natural infection.
Design:Prospective cohort.
Setting:Nursing home.
Participants:SARS-CoV-2–infected nursing home residents.
Methods:A convenience sample of 14 SARS-CoV-2–infected nursing home residents, enrolled 4–13 days after real-time reverse transcription polymerase chain reaction diagnosis, were followed for 42 days. After diagnosis, plasma SARS-CoV-2–specific pan-Immunoglobulin (Ig), IgG, IgA, IgM, and neutralizing antibodies were measured at 5 time points, and GCF SARS-CoV-2–specific IgG and IgA were measured at 4 time points.
Results:All participants demonstrated immune responses to SARS-CoV-2 infection. Among 12 phlebotomized participants, plasma was positive for pan-Ig and IgG in all 12 participants. Neutralizing antibodies were positive in 11 participants; IgM was positive in 10 participants, and IgA was positive in 9 participants. Among 14 participants with GCF specimens, GCF was positive for IgG in 13 participants and for IgA in 12 participants. Immunoglobulin responses in plasma and GCF had similar kinetics; median times to peak antibody response were similar across specimen types (4 weeks for IgG; 3 weeks for IgA). Participants with pan-Ig, IgG, and IgA detected in plasma and GCF IgG remained positive throughout this evaluation, 46–55 days after diagnosis. All participants were viral-culture negative by the first detection of antibodies.
Conclusions:Nursing home residents had detectable SARS-CoV-2 antibodies in plasma and GCF after infection. Kinetics of antibodies detected in GCF mirrored those from plasma. Noninvasive GCF may be useful for detecting and monitoring immunologic responses in populations unable or unwilling to be phlebotomized.

Introduction
Pest pressure is the inevitable, ubiquitous factor in evolution which makes for an apparently pointless multiplicity of species in all areas in which it has time to operate.
(Gillett 1962)At a symposium 44 years ago, J. B. Gillett proposed the Theory of Pest Pressure, whereby plant pathogens and pests were responsible for the genesis and maintenance of high plant diversity in tropical forests and other high-diversity systems. In his conclusion to the paper produced from that talk he hoped that this ‘new theory may be useful in stimulating discussion and research’ on the roles of pests and pathogens as a force in plant diversity. Apparently his theory has been useful, as much has happened in the last four decades to explore and expand on his idea. My aim is to review key research on the effects of plant pathogens in tropical forests since Gillett's seminal paper with emphasis on the special case of his Theory of Pest Pressure known as the Janzen–Connell Hypothesis. I will also suggest critical areas that need to be explored as we continue to discuss and research the role of pathogens in the maintenance of species diversity in tropical forests.
‘Parcere subiectis et debellare superbos’
Gillett (1962) paraphrased Virgil's famous formula for the greatness of Rome, ‘Spare the lowly and conquer the haughty’ (AeneidVI: 853), when he first introduced the idea that if plant pests have a greater impact on more common species than on rare species, the rare species should then increase in relative frequency, providing a mechanism for the maintenance of diversity in species-rich communities.

Scope of the problem
Ear pain (otalgia) is a common emergency department (ED) complaint. It prompts over 30 million physician visits per year. By the third year of life, 80% of the population will have complained of otalgia at least once. Though many conditions may cause ear pain, otitis media (OM) is by far the most common diagnosis, especially in the pediatric population. The potential for serious causes of otalgia, such as malignant otitis externa (OE) and mastoiditis, underscore the need for early and accurate diagnosis and treatment.
Anatomic essentials
The anatomic ear may be divided into three distinct sections: external, middle and inner (Figure 19.1). The external ear, consisting of the auricle (pinna) and the external auditory canal (EAC), originates at the pinna and ends at the tympanic membrane (TM). The middle ear is an air-containing cavity in the petrous temporal bone that houses three auditory ossicles: the malleus, incus and stapes. Though the middle ear is separated from the outer ear by the TM, it connects anteriorly with the nasopharynx via the eustachian tube, and posteriorly with the mastoid air cells. The inner ear includes the cochlea, which contains the auditory receptors, and vestibular labyrinth, which contains the balance receptors. Sensory innervation of the ear is derived from branches of cranial nerves (CNs) V, VII, IX and X as well as I, II and III.
Primary otalgia (Table 19.1) is ear pain that results from structures directly within or adjacent to the anatomic ear.

Datronia caperata, a basidiomycete fungus, is one of the dominant polypore species found in neotropical mangrove forest fragments, where it is locally specialized on Laguncularia racemosa. We examined the genetic structure of D. caperata populations from four Panamanian mangrove forests using AFLP markers. Using five primer pair combinations, 145 loci were detected, 98.6% of which were polymorphic. Each of the populations showed a high degree of genetic diversity (Nei's h ranging from 0.146 to 0.223). Results from minimum spanning trees and Mantel tests showed little evidence for small-scale spatial structure within sites. A significant amount of total genetic variation was partitioned among populations (ΦST=0.21) separated by 10s to 100s of km, a considerably greater amount than has been detected in other mushroom and wood-decaying fungi sampled at equal or greater geographic distances. These results suggest that despite production of copious basidiospores capable of long distance dispersal, some homobasidiomycete fungi may be susceptible to genetic isolation due to habitat fragmentation.

Fungal endophytes have been found in every plant species examined to date and appear to be important, but largely unquantified, components of fungal biodiversity. Endophytes are especially little known in tropical forest trees, where their abundance and diversity are thought to be greatest. Here, we explore the occurrence of endophytes in a broad diversity of woody, angiospermous taxa in a lowland, moist tropical forest in central Panamá. We use similarity indices to assess host preference and spatial heterogeneity of endophytes associated with two co-occurring, but distantly related, understorey tree species in two sites of that forest, and assess the utility of indices based on frequencies of morphospecies occurrence (Morisita-Horn index) and on presence-absence data (Sørensen’s index). We suggest that our understanding of fungal diversity will be enhanced by exploring ecological patterns underlying endophyte occurrence in host species, and discuss methods for assessing the proportion of fungal biodiversity represented by tropical endophytes.

Growth and survival of aerial roots of hemiepiphytes in a lower montane tropical moist forest in Panama

Hemiepiphytic plants grow for part of their life as true epiphytes, then become terrestrial through the production of aerial roots that grow from the canopy to the ground. Long-term measurement of growth, dieback and mortality of aerial roots of hemiepiphytic plants in a lower montane moist tropical forest in western Panama was used to elucidate life-history strategies of hemiepiphytes from two families. The fates of 156 aerial roots of five species of Clusiaceae and Araceae were followed for 10 mo. Some roots were cut to experimentally study the effect of injury on resprouting and survival. Aerial roots of Araceae grew more than twice as fast as those of Clusiaceae but had a much greater mortality rate. Roots of both families grew much faster during the wet than dry season. Even for the fastest growing roots, growth and survival models suggest that only 18% of Araceae roots were likely to survive long enough to reach the ground from a branch 10 m high, whereas 87% of roots of Clusiaceae were likely to do so. This suggests that only those Araceae hemiepiphytes that produce a large number of aerial roots or are located close to the ground are likely to reach the soil.
